Title: “Neural Stem Cell Fate Specification in the Development of the Human Brain and Glioblastoma”

Abstract: The developing human brain is composed of many cell types and subtypes that give rise to the complex functions of human perception, cognition, and behavior. We use single-cell RNA sequencing to create an atlas of cell types during human brain development, with a special focus on the cortex and different areas within the cortex. These profiles have identified transcriptional patterns that are essential to brain and cortical patterning, and highlight spatiotemporal regulation patterns including dynamics changes across cell maturation and differentiation. This ground-truth dataset is essential to evaluating the performance of in vitro models, and these comparisons identify both similarities and differences between cortical organoid models and the developing human cortex. Moreover, comparisons of single-cell profiles in development to glioblastoma show the reactivation of developmental cell types in the cancer context, and suggest a role for outer radial glia-like cells in cancer progression and invasion.
